Macrophage responses to CAR T cell produced cytokines across Autoimmune Diseases may confer differential risk among indications

Evaluated primary cell-derived macrophages from healthy donors alongside systemic sclerosis (SSc) and idiopathic inflammatory myositis (IIM) patients following exposure to pro-inflammatory T-cell cytokines.
Monocyte-derived macrophages (MDMs) from SSc patients appeared to upregulate pro-inflammatory markers at lower concentrations of IFNγ and TNFα than those from IIM patients, suggesting potential indication-dependent sensitivity differences.
Observed variations in myeloid responsiveness may offer insight into potential differences in CRS and ICANS patterns across distinct autoimmune disease populations receiving CD19 CAR T-cell therapy.
